Tirzepatide is a synthetic peptide studied for its activity at two incretin receptors: glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1). Most earlier incretin compounds act mainly at GLP-1. Tirzepatide was designed to engage both.
What the literature describes
Published pharmacology papers describe tirzepatide as a biased dual agonist. In receptor assays it activates GLP-1 and GIP receptors, with a longer circulating half-life than native incretin hormones because of a fatty-acid side chain that binds albumin. That pharmacokinetic feature is why research protocols often use infrequent dosing intervals compared with unmodified peptides.
Preclinical and clinical literature has focused on glucose handling, appetite signalling, and body-composition endpoints. Those papers are drug-development studies. They are not a licence to use a research vial as a medicine.
How it differs from semaglutide
Semaglutide is primarily a GLP-1 receptor agonist. Tirzepatide adds GIP receptor activity. Head-to-head papers compare the two on metabolic endpoints, but the compounds are not interchangeable in a research protocol. Sequence, receptor profile, and dose are different.
